จากกรุงเทพฯ วิ่งไปตามทางหลวงแผ่นดินหมายเลข 4 (เพชรเกษม) ผ่านจังหวัดเพชรบุรี จนถึงอำเภอปราณบุรี แล้วเลี้ยวซ้ายที่บริเวณสี่แยกปราณบุรีไปตามถนนสายปากน้ำปราณบุรี ประมาณ 4 กิโลเมตร แล้วจึงเลี้ยวขวาไปตามถนน รพช. อีก 31 กิโลเมตร ก็จะถึงที่ทำการอุทยานแห่งชาติ อีกเส้นทางหนึ่ง จากกรุงเทพฯ วิ่งไปตามทางหลวงแผ่นดินหมายเลข 4 (เพชรเกษม) จนถึงหลักกิโลเมตรที่ 286.5 ใกล้บ้านสำโรง ก่อนถึงอำเภอกุยบุรีประมาณ 6 กิโลเมตร แล้วเลี้ยวซ้ายไปอีก 14 กิโลเมตร ก็จะถึงที่ทำการอุทยานแห่งชาติ
